I would like to selectively enable/disable a simple redirect from a subdomain to a third-party domain. Is there a way to programmatically do this beyond committing a comment in the _redirect file?

Unfortunately not, you’ll need to rebuild. However, if nothing except the _redirect file changes, it should cache and build pretty quickly!
